Extracellular Superoxide Dismutase (EC-SOD) Regulates Gene Methylation and Cardiac Fibrosis During Chronic Hypoxic Stress

چکیده

Chronic hypoxic stress induces epigenetic modifications mainly DNA methylation in cardiac fibroblasts, inactivating tumor suppressor genes (RASSF1A) and activating kinases (ERK1/2) leading to fibroblast proliferation fibrosis. The Ras/ERK signaling pathway is an intracellular signal transduction critically involved proliferation. RASSF1A functions through its effect on downstream ERK1/2. antioxidant enzyme, extracellular superoxide dismutase (EC-SOD), decreases oxidative from chronic hypoxia, but effects these changes have not been fully explored. To test our hypothesis, we used in-vitro model: wild-type C57B6 male mice (WT) transgenic males with extra copy of human hEC-SOD (TG). studied animals were housed hypoxia (10% O 2 ) for 21 days. right ventricular tissue was fibrosis markers using RT-PCR Western blot analyses. Primary C57BL6 mouse culture study the model, RASSF-1 expression methylation, relation Our findings showed a significant increase markers: Collagen 1, alpha smooth muscle actin (ASMA), SNAIL, WT as compared TG group ( p < 0.05). enzymes (DNMT 1&3b) significantly increased 0.001). lower ERK1/2 higher Use SiRNA block gene murine led Methylation promoter region reduced (0.59 vs. 0.75, respectively). Based findings, can speculate that EC-SOD attenuates alleviate induced by hypoxia.

Human extracellular superoxide dismutase (EC-SOD) expression in transgenic chicken

Extracellular superoxide dismutase (EC-SOD) is a metalloprotein and functions as an antioxidant enzyme. In this study, we used lentiviral vectors to generate transgenic chickens that express the human EC-SOD gene. The recombinant lentiviruses were injected into the subgerminal cavity of freshly laid eggs. Subsequently, the embryos were incubated to hatch using phases II and III of the surrogate...

Extracellular Superoxide Dismutase (ec-sod) as a Protective Factor for Risk of Chronic Obstructive Pulmonary Disease

WINNING ABSTRACT: Tobacco smoke contains a high concentration of oxidants and is the primary cause of chronic obstructive pulmonary disease (COPD). Extracellular superoxide dismutase (EC-SOD) is the major antioxidant enzyme in the extracellular space of the lung and is part of the lung defense against these oxidants. We hypothesized that EC-SOD is a risk factor for COPD. We found that EC-SOD pl...

Leukocyte-derived extracellular superoxide dismutase does not contribute to airspace EC-SOD after interstitial pulmonary injury.

The antioxidant enzyme extracellular superoxide dismutase (EC-SOD) is abundant in the lung and is known to limit inflammation and fibrosis following numerous pulmonary insults. Previous studies have reported a loss of full-length EC-SOD from the pulmonary parenchyma with accumulation of proteolyzed EC-SOD in the airspace after an interstitial lung injury. However, following airspace only inflam...

GroESL protects superoxide dismutase (SOD)— Deficient cells against oxidative stress and is a chaperone for SOD

Superoxide dismutase (SOD)-deficient Escherichia coli OX326A cells are protected against chemically-induced oxidative stress by expression of the chaperonin GroESL. This protection is equivalent to expression of superoxide dismutase even though GroESL has no inherent SOD activity. Co-overexpression of GroESL and SOD in the same cells results in higher protein yields of SOD and greater metallati...
